Responsive Sensory Evaluation to Develop Flexible Taste-Masked Paediatric Primaquine Tablets against Malaria for Low-Resource Settings

Primaquine is an important antimalarial drug for malaria transmission blocking and radical cure, but it not currently available in child-friendly formulations appropriate doses. Adult-strength tablets are often crushed dissolved water to obtain the required dose, which exposes drug’s bitter taste. As part of developing paediatric primaquine (DPP) project, this study adopted a responsive sensory pharmaceutics approach by integrating real-time formulation development pre-clinical taste assessment develop palatable, flavour-infused tablets. A design experiment (DoE) was used screen different taste-masking agents excipient blends with trained, expert assessors, quinine hydrochloride as model tastant. The efficacy selected prototype validated naïve assessors using highest 15 mg dose. mean bitterness intensity rating, measured on discrete 11-point scale, halved from 7.04 unflavoured control 2.74–3.70 blends. Sucralose had biggest impact suppression improving palatability. Two flavouring systems have been developed, their acceptability patients will be assessed upcoming validation field clinical trials Africa.
